The Bayliss lab, at the University of Virginia School of Medicine Department of Pharmacology, seeks candidates for a Lab Specialist Senior. The Bayliss Laboratory studies the cellular, molecular and biophysical mechanisms of ion channel function and modulation. A particular interest is in understanding the fundamental properties of large pore channels (e.g., pannexin channels), and how their contribution to lung inflammation and blood pressure regulation. We utilize mouse models and primary cell culture to test biological mechanisms. The candidate will work closely with other members of the laboratory, with primary duties including, but not limited to: Molecular biology: Cloning, mutagenesis, sequencing, and cataloging channel constructs in various vectors that can be used for expression in mammalian cells (for functional studies) and in insect (Sf9) cells for protein purification. Maintaining freezer stocks and cell lines. Cell culture: Maintain and propagate cell lines for channel expression. Maintaining freezer stocks and cell lines. Channel biochemistry: Perform Western blotting of channels expressed in mammalian cells. Use baculovirus vectors for large-scale production of channels for protein biochemistry and structural studies. Prepare proteoliposomes for functional analyses of single channel currents, and large molecule permeation. Cell isolation: Isolate primary cells from mouse lines with channel mutations, including immune cells and vascular smooth muscle cells. Prepare primary cells for recording and other biochemical and functional assays. Experimental design and analysis: Contribute to planning and execution of experimental protocols, data analysis and manuscript preparation. The candidate will be required to present this data in lab meetings. Proficiency in Microsoft Word, Excel and PowerPoint is required. Experience with statistical analysis and graphing software is a plus. Lab Inventory and Stocking: The candidate will be responsible for creating lab inventories of available reagents, stocking the lab, ordering supplies and prepare solutions and buffers. Optimization and maintenance of lab protocols for use by all lab members: The candidate will optimize and archive lab protocols for later use and will be required to keep a detailed and accurate lab notebook that others can follow. Collaboration and training: Importantly, this lab will consist of multiple trainees at all levels, from undergraduate to postdoctoral fellows to research faculty, and so the ideal candidate must be willing to work as a team, create an inclusive environment and be willing to train new lab members on existing protocols in the lab. MINIMUM REQUIREMENTS Education: Bachelor's degree. Experience: 6 years of experience. 10 years of lab-related experience accepted in lieu of degree or Master's degree plus 4 years lab-related experience. PHYSICAL DEMANDS This position can spend extensive periods of time standing and squatting while conducting experiments or caring for animals. Also requires keyboarding and other bench work and animal procedures requiring finer finger manipulation. Salary will be commensurate with education and experience.
